Luciferase is a general term for enzymes that can produce bioluminescence in nature. The most representative types of luciferase are those from Firefly and Renilla. Luciferase catalyzes the conversion of specific luciferin substrates to oxyluciferin and emits intense light.
HanBio provides luciferase and dual luciferase detection kits to help you operate experiments easily and efficiently.

Luciferase (Firefly) Reporter Assay Kit
Firefly luciferase (fluc) can catalyze the bioluminescence reaction of luciferin. The reaction process is shown in the following figure.
The assay kit used fluorescein as a substrate to detect the activity of firefly reporter gene. This method has the advantages of simple operation, fast detection and high sensitivity.
Dual Luciferase Reporter Assay Kit
Firefly luciferase (fluc) and renilla luciferase (renilla luciferase, rluc) can catalyze the bioluminescence reaction of luciferin and coelenterazine, respectively, as shown in the following figure.
In the dual-luciferase Reaction, the fLuc signal was detected by Luciferase Reaction Reagent I. Subsequently, Luciferase Reaction Reagent II was added directly. The signal of fLuc was quenched and the rLuc signal was generated at the same time. This method has the advantages of simple operation, fast detection and high sensitivity.

The kit typically includes luciferase assay buffer, substrate solution, and any necessary reagents for the detection process. Detailed instructions and a protocol booklet are also included.
The components should be stored as per the instructions provided in the kit manual, usually at -20°C for long-term storage. Some components may need to be stored at 4°C after reconstitution.
The basic procedure involves preparing the cell lysates or samples, adding the luciferase substrate, and measuring the luminescence using a luminometer. Detailed step-by-step instructions are provided in the kit manual.
The assay is relatively quick and can typically be completed within 30-60 minutes, depending on the number of samples and the specific protocol used.
The kit is highly sensitive and capable of detecting low levels of luciferase activity, making it suitable for a wide range of applications, including those requiring high sensitivity.
