Gene synthesis does not require a template and involves amplifying DNA fragments in vitro using overlapping short primers, which range in length from 100bp to 10kb. This method is suitable for obtaining various target genes that are difficult to access, non-natural sequences, amino acid mutations, or rare codon substitutions, allowing for the fastest acquisition of the desired sequence with 100% accuracy.
